| Abstract |
| The structure and function of skeletal muscle tissue, like other tissues, are disrupted in conditions of a high-fat diet, which may underlie many obesity disorders. Aerobic exercise and medicinal herbs can reduce the negative effects of a high-fat diet on muscle tissue. Therefore, the aim of the present study was to determine the effect of aerobic exercise and anti-obesity herbal supplement on the expression of genes in the intrinsic pathway of apoptosis in the skeletal muscle tissue of male rats. In an experimental study, 30 male Wistar rats were selected as subjects and randomly divided into five groups: 1- Normal diet control group, 2- High-fat diet control group, 3- High-fat diet-aerobic exercise group, 4- High-fat diet-anti-obesity herbal supplement extract group, and 5- High-fat diet-aerobic exercise-anti-obesity herbal supplement extract group. The aerobic training program consisted of four weeks of moderate-intensity treadmill running. The anti-obesity supplement extract was also administered to the subjects by gavage. After the intervention, the rats were sacrificed and skeletal muscle tissue was removed to measure the expression of BAX, BCL-2, and Caspase-3 genes. The results showed that obesity induction significantly increased BAX and Caspase-3 gene expression, while BCL-2 gene expression was considerably decreased. Aerobic exercise and anti-obesity supplementation significantly decreased BAX and Caspase-3 gene expression and greatly increased the expression of BCL-2 genes. The highest changes were observed in the combination of aerobic exercise and anti-obesity supplement group. Based on the results obtained, it is suggested that the simultaneous use of aerobic exercise with an anti-obesity supplement is a beneficial strategy for modulating the negative effects of a high-fat diet on skeletal muscle tissue by regulating the expression of genes of the intrinsic apoptosis pathway.
